Glysosidases in Chlorella secretome

A one day NC64a culture was pelleted, resuspended in 30ml of MBBM w/0.5% sucrose and 1ml tetracycline, incubated overnight and pelleted. The supernatant (brought to 0.05% in sodium azide) was assayed (below) for glycosidases. The numbers are microliters of supernatant added to a 100 microliter substrate solution. Incubation was for 35 minutes at 37C

D-NAc-beta-glucosaminidase is a relatively stable enzyme and could be assayed at higher temperatures (50C). There is enough enzyme so that it should be detectable in culture supernatants w/o concentration.
